In plain words

The One has been your Master and Guardian all along — keep saying the Naam of that One.

Nanak says: this teaching is for you. If you listen and take it in, your pain will go.

Listening to what is true can ease the ache inside.
The deeper layer

Listen to the teaching, and the ache inside begins to ease.

Tumray prabh hotay su-aamee means the One has been your Master and Guardian all along. Jaapahu jaap means keep repeating and holding that Name. Updays karat is the teaching, offered with love.

Then comes the promise: ja-o sunhu ta-o jaa-ay santaap, "if you listen, the suffering goes away." This is the healing power of listening, which Guru Nanak also praises in Japji Sahib. Think of a friend who keeps saying, "Drink some water, you'll feel better." It only helps when you actually listen. Real listening eases the ache.
